Kof was originally pronounced with a k sound produced from the throat, almost like a q. (This sound is still present in Arabic.) In fact, the Hebrew letter Kof is often transliterated as q in English, such as in the word Qumran. Now, however, Kof is pronounced as k, as in the words Korah, Cain, or kill. In Hebrew today there is no difference between the pronunciation of Kof and Kaf with a dagesh.

Kof has the Gematria (numerical value) of 100.
